Acı kavun (Ecballium elaterium L. 1758)'un sazan (Cyprinus carpio L. 1758) balıkları üzerindeki mikronükleus oluşumuna ve kan hücre morfolojilerine etkileri

Abstract

Yüzyıllardır hastalıkların tedavisinde diğer tedavi seçeneklerinin yanı sıra bitkiler ve bitkisel ilaçlardan da yararlanılmaktadır. Bu bitkilerden birisi olan acı kavun, acı kelek ve şeytan keleği de denilen Ecballium elaterium bitkisi sinüzit, egzama gibi hastalıkların tedavisinde kullanılmaktadır. Kullanıldığında tedavi edici özelliğinin yan ısıra, insanlar üzerinde ciddi biyolojik hasarlar da bıraktığı görülmektedir. Bu çalışmada, acı kavun meyvelerinden elde edilen özsuyun sazan balıklarının hücre morfolojilerine etkisinin olup olmadığı ve mikronükleus oluşumuna etkisinin belirlenmesi amaçlanmıştır. Uygulama 3 doz ve 1 kontrol grubu olmak üzere 4 grup üzerinde yapıldı. Üç farklı gruba ait akvaryumlara sırasıyla 100, 200 ve 300 ppm dozlarda acı kavun özsuyu 72 saat uygulandı. Balıklardan kan yayma preparatları hazırlandı. Hazırlanan preparatlar mikroskop altında taranarak kan hücrelerinin morfolojilerinde meydana gelen değişiklikler ve mikronükleus oluşmuş eritrositler tespit edildi. Deney grubuna ait preparatlarda yapılan taramalarda tespit edilen hücreler ile kontrol grubundaki hücrelerin morfolojileri arasında farklılıklar olduğu gözlendi. Sonuç olarak acı kavun özsuyunun sazan balığı kan hücrelerinde bölünme esnasında mikronükleus oluşumuna neden olduğu ve kan hücrelerinin morfolojilerinde bazı bozukluklara neden olduğu tespit edildi. Bu veriler ile acı kavun özsuyunun genotoksisiteye neden olduğu düşünülmektedir.

Plants and plant-based medications are being utilized. One of these plants, bitter melon, Ecballium elaterium and also referred to as the bitter unripe melon and the squirting cucumber. It is also seen that along side its therapeutic properties when used, it leaves serious biological damage on human. This study aims to see whether the juice acquired from these bitter melon fruits have any effect on the morphologies of the cells of carp and its effect on micronucleus formation. It was administered in 4 groups, of which 3 were dosage groups and 1 was a control group. The bitter melon juice was administered over the course of 72 hours in 100, 200, and 300 ppm doses to the aquariums belonging to the three different dosage groups. Blood smear preparations were made from the fish. By scanning the prepared preparations under a microscope, the changes that occurred in the morphologies of the blood cells of the fish and the blood cells in which micronucleus formed were identified. In the preparations belonging to the control group, it was seen that there were differences between the cells identified in the scans and the morphologies of the cells in the control group. As a result, it was determined that bitter melon juice leads to micronucleus formation during the division in blood cells of carp and that it leads to some disorders in the morphologies of blood cells. With these data, it is thought that bitter melon juice causes genotoxicity.
